Design Notes
First, we divided the lacZ coding sequence into two parts, the first 122 amino acids, and the last 123 amino acids.
Then, we divided the intein coding sequence into two parts, the first 206 amino acids, and the last 387 amino acids.
Most importantly, we combine the sequences in the following order: the first 122 amino acids of lacZ, the first 206 amino acids of the intein, the hera coding sequence, the last 123 amino acids of lacZ, and the last 387 amino acids of the intein.
